Clinical significance of regulation of prolyl hydroxylases 3 by Siah2 in non-small cell lung cancer
Abstract:Objective To investigate the expression of Siah2(seven in absentia2) and prolyl hydroxylase 3(PHD3) in non-small cell lung cancer ( NSCLC) ,and to elucidate their clinical significance .Methods The mRNA expression levels of Siah2 and PHD3 were detected by RT-PCR in 89 cases of NSCLC and adjacent normal lung tissues , and their protein expression levels were detected by Western Blot .Results The expression levels of Siah 2 mRNA and protein in tumor tissues were significantly higher than those in adjacent tissues ( P <0.05).The expression levels of PHD3 mRNA in tumor tissues were obviously higher than those in adjacent tissues , however , the expression levels of PHD 3 protein in tumor tissues were significantly lower than those in adjacent tissues ( P <0.05).The expression of Siah2 protein was negatively correlated to that of PHD3 protein( r =-0.735, P <0.05),however,the expression of Siah2 protein was positively related with that of Siah 2 mRNA( r =0.821, P <0.05).Conclusion Siah2 may play a role in pathogenesis of NSCLC by regulating PHD 3 expression levels.
