Study on the relationship between the expression of apoptosis-related bcl-2 protein as well as mRNA of luteined granulosa cells and ovarian reactivity
Abstract:Objective To investigate the correlation between the expression of apoptosis-related bcl-2 bcl-2 protein as wellas mRNA of human luteined granulosa cells ( GCs) and ovarian reactivity .Methods Twenty -eight infertility cases receiving IVF-ET were enrolled in the study .The experiment was divided into three groups according to ovarian follicle number ,low reactivity group , median reactivity group and high reactivity group .The expression of bcl-2 protein of GCs was detected by immunocytochemistry;expression of bcl-2 mRNA of GCs was detected by RT-PCR.Finally the correlation between the expression of apoptosis-related bcl-2 protein as well as mRNA of human luteined granulosa cells ( GCs ) and ovarian reactivity was analyzed.Results The isolated and purified GCs were cultured and adhered well within 24 hours. Immuocytochemistry examination showed that the HSCORE scoring of bcl-2 protein ( HSCbcl-2) was gradually increased with the increase of number of retrieved oocytes , and HSCbcl-2 was positively correlated to number of retrieved oocytes ( P <0.01).There were significant differences in HSCbcl-2 of GCs among the three groups ( P <0.05).RT-PCR showed that the expression of bcl-2 mRNA of GCs was negatively related with subject ’ s age ( P <0.01),however,which was positively correlated to the number of retrieved oocytes ( P <0.05).There were significant differences in the expression levels of bcl-2 mRNA of GCs among the three groups ( P <0.05).Conclusion The expression of apoptosis-related bcl-2 protein and mRNA of luteined granulosa cells is closely correlated to ovarian reactivity .
