The clinical significance of expression and DNA binding activity of hypoxia-inducible factor 1-αin non-small cell lung cancer
Abstract:Objective To observe the dynamic changes of expression and DNA binding activity of hypoxia -inducible factor 1-αin non-small cell lung cancer ( NSCLC ) , and to investigate the correlation between the changes and tumor pathological characteristics .Methods Eighty-nine cases of NSCLC tissues and adjacent normal lung tissue specimens were collected,RT-PCR was used to detect the expression levels of HIF-1αmRNA,and Western Blot was used to determine the expression levels of HIF-1αprotein.EMSA assay was used to examine DNA binding activity of HIF-1α.The correlation between the expression levels and the clinical pathological features of tumors was analyzed .Results The expression levels of HIF-1αmRNA and protein in tumor tissues were significantly higher than those in adjacent tissues ( P <0.05).Their expression levels were correlated with tumor staging and lymph node metastasis ( P <0.05).HIF-1αDNA binding activity was significantly increased,as compared with that in adjacent normal tissue ( P <0.05).Conclusion The overexpression of HIF-1αand enhancement of transcription activity are closely correlated to pathogenesis of NSCLC ,which can be regarded as a target in inhibiting pathogenesis of NSCLC .
