Effects of speranskiat uberculataex tract on proil feration of human hepatocellular carcinoma SMMC-7721 cells

Abstract：Objective To observe the effect of speranskia tuberculata extract on proliferation of human hepatocellular carcinoma SMMC-7721 cells, and to explore its molecular mechanism .Methods The effects of speranskia tuberculata extract on proliferation of human hepatocellular carcinoma SMMC -7721 cells were detected by MTT,and the expression levels of β-catenin and cyclin D1 protein in SMMC-7721 cells were determined by immunohistochemistry method.Results The inhibition rate on proliferation of SMMC-7721 cells treated by speranskia tuberculata extract at the concentration of 6.25~100μg/ml was significantly increased ,as compared with that in control group ( P <0.05),moreover,with the increase of concentration of speranskia tuberculata extract,the inhibition rate was gradually increased .The IC50 of speranskia tuberculata extract on SMMC-7721 cells was 40.91 μg/ml.The expression levels of catenin and cyclin D 1 in SMMC-7721 cells treated by speranskia tuberculata extract at the concentration of 40.91μg/ml were significantly lower than those in control group ( P <0.05 ).Conclusion The speranskia tuberculata extract has inhibition effect on proliferation of human hepatocellular carcinoma SMMC-7721 cells, and its action mechanism may be related with down-regulating the expression of β-catenin and cyclin D1 protein.
