Effect of celecoxib on the expression of IL-8 and MCP-1 in peripheral blood of rats with acute lung injury
Abstract:Objective To investigate the effects of celecoxib on the expressions of peripheral blood chemokines-interlukine 8 ( IL-8) and monocyte chemoattractant protein ( MCP-1 ) in rats with acute lung injury induced by lipopolysaccharide ( LPS) .Methods Forty-five rats were randomly divided into three groups:control group,LPS group and celecoxib treatment group ,with 15 rats in each group.The rats in control group were given 0.9%sodium chloride solution ,the animal models with acute lung injury were established by intravenous injection LPS(5 mg/kg) and the rats in celecoxib treatment group were given celecoxib (20mg /kg) at 30min after LPS induction,then the rats were sacrificed after 3h.The expression levels of IL-8 and MCP-1 in peripheral blood of rats were detected by ELISA .Results There were significant differences in the expression levels of IL-8 and MCP-1 between LPS group and control group ( P <0.05).There were significant differences in the expression levels of IL-8 and MCP-1 between celecoxib treatment group and LPS group ( P <0.05).Conclusion Celecoxib can improve the reaction extent of rats to acute lung injury by inhibiting the expression levels of IL -8 and MCP-1.
