Study on the extraction, purification and antidepressant activity of total saponins extracted from Sanchi leaf
Abstract：Objective To study the optimum extracting technology and its antidepressant activity of the total saponins extracted from Sanchi leaf .Methods The orthogonal experiment was adopted to observe the effects of ethanol concentration,extraction temperature,ratio of solid to liquid and extraction time on the total saponins extraction efficiency , and the total saponins of Sanchi leaf were separated and purified by D-101 porous resin absorption and step-sedimentation with acetone-ether.The antidepressant effects of the total saponins of Sanchi leaf were evaluated by forced swimming test and the tail suspension test in mice .Results The optimum conditions of extracting total saponins from Sanchi leaf were extraction time:1.5h,ethanol concentration:60%,solid /lipuid ratio:1∶10 and extraction temperature:80℃.As compared with total saponins from Sanchi leaf like the control drug-fluoxetine could significantly shorten immobility time of swimming and tail suspension in mice .Conclusion The optimum extraction condition and purification method are very useful for further development of industrial process.The total saponins extracted from Sanchi leaf has effective antidepressant activity .
