Optimization of Ultrasonic-assisted Extraction Technology and Antioxidant Activity of Polyphenols from Raphanus Sativus Leaves

Abstract:In order to improve the utilization rate of Raphanus sativus leaves resources and explore their potential application value,the ultrasonic-assisted extraction technology for polyphenols from Raphanus sativus leaves was explored and its antioxidant activity was evaluated.By introducing Box-Behnken response surface method on the basis of single factor experiment,the ultrasonic-assisted extraction process of polyphenols from Raphanus sativus leaves was optimized,and its antioxidant capacity was further verified by the evaluation of in vitro experiments.The results showed that under the optimum conditions of ultrasonic temperature of 68℃,liquid-solid ratio of 32 mL/g,ethanol volume fraction of 63%and ultrasonic time of 31 min,the extraction yield of polyphenols from Raphanus sativus leaves was 60.84mg/g,and the relative error was as low as 0.73%,which fully confirmed the effectiveness and feasibility of response surface analysis to optimize the extraction process.Polyphenols from Raphanus sativus leaves showed certain antioxidant properties,and the half maximum inhibitory concentration(IC50)of DPPH and O-2 radical were 78.91mg/L and 403.55mg/L,respectively,which provided reference for further research and development of antioxidants from natural plants.
