Abstract:Aiming at the problems such as the unclear pattern of the selenium accamulation and the immature extraction technology of selenium-containing organic compounds in vegetable-used Astragalus sinicus japonica before budbreak.Using the organic selenium extraction rate as an indicator, response surface analysis was used to investigate the factors affecting the extraction of organic selenium compounds in selenium-enriched vegetable-used Astragalus sinicus powder, and to optimize the extraction process of the main selenium-containing organic compounds. The experiment showed that the organic selenium content of Astragalus sinicus accounted for 92.49%of the total selenium,of which alkali soluble protein selenium accounted for 69.45%of the total selenium; The optimal process conditions for the extraction of organic selenium were:the material concentration of sodium hydroxide solution was 0.15mol/L,the dosage of sodium hydroxide solution was 20 times the volume of the sample, the extraction time was 2h,and the number of extractions was 2 times. The results showed that the selenium in Astragalus sinicus was mainly in the organic bound state, and the organic bound state selenium was best extracted in alkaline solution. When extracting the organic selenium in Astragalus sinicus, the acidity and alkalinity of the extraction solution, the dosage of extraction solution, the extraction time and the number of times of extraction and other factors should be considered. The results can provide a scientific basis for the further development and utilization of Astragalus sinicus for vegetable use.
