The Ameliorating Effect of Deer Skin Gelatin Polypeptide-iron Chelate against Iron Deficiency Anemia in Young Mice

Abstract:In this study,a deer skin gelatin peptide-Fe(DSGP-Fe)chelate was prepared using deer skin gelatin as the raw material,which was subjected to proteolytic hydrolysis and ferrous chloride(FeCl2-4H2O)treatment,and the preliminary pharmacological study was carried out on its blood-supplementing effect.The structure was analyzed using Fourier infrared spectroscopy and scanning electron microscopy;an iron-deficiency anemia model was established by a combination of feeding an iron-deficient diet and bloodletting.After the model was successfully established,the corresponding medication was given until the Hb value was higher than 110 g/L.At the end of the experiment,theorgan indexes,iron metabolism-related serum factor indexes,liver injury-related indexes,and oxidative stress indexes of the mice were measured.Characterization results show that DSGP-Fe had a loose bulk structure,in which Fe2+mainlybound to the chelating active groups such as amino and carboxyl groups in deer skin gelatin peptide(DSGP)to form DSGP-Fe.Ferrous succinate tablets,DSGP and DSGP-Fe increased the IDA group's body weight(42.3,35.7,41.8 g),splenic index(3.58,3.91,3.63 mg/g)and hepatic index(2.31%,2.02%,2.28%),SI(32.52,27.35,35.47 mg/L),SF(34.34,32.21,36.34 ng/L)and TSAT(7.49%,7.25%,7.32%)levels,decreased their TIBC(4.34,3.77,4.84 mg/L)levels,raised their GSH(13.34,11.34,14.65 pmol/g prot)and SOD(209.35,193.52,216.58 U/mg prot)activities,and reduced ALT(27.34,20.77,20.34 U/L),AST(61.34,50.21,50.34 U/L),and MDA(1.54,1.67,1.52 nmol/mg prot)contents.In conclusion,DSGP-Fe exerted significant effects on iron supplementation,regulation of iron metabolism,and enhancement of anti-oxidative stress capacity in young mice with iron-deficiency anemia.Therefore,DSGP-Fe can be used as a new potential source of iron,and provides a new idea for the development of iron fortifiers from deer skin gelatin.
