Inhibitory Effects of Mulberry Leaf Leachates on Proliferation of MDA-MB-231 Cells and Their Survivin Protein Expression

Abstract:The aim of this paper was to study the effects of mulberry leaf leachates on the proliferation of MDA-MB-231 breast cancer cells and antiapoptotic protein survivin.MDA-MB-231 cells were cultured in vitro,and the morphological changes were examined under an inverted microscope.The effect of the mulberry leaf leachates on the cell proliferation was detected by the cell counting kit-8 (CCK8) method.The influence of the mulberry leafleachate on apoptosis and changes in the cell cycle of MDA-MB-231 cells were analyzed by flow cytometry.Finally,the effect of the mulberry leaf leachates on the expression of the survivin protein was detected by westem blotting.The results showed that the leachate of mulberry leaves significantly changed the growth and morphology of MDA-MB-231 cells:cell proliferation was inhibited,and the number of apoptotic cells increased significantly.After thetreatment with the mulberry leaf leachate,the number of MDA-MB-231 cells at the G0/G 1 transition was significantly higher than that of the normal group (p < 0.05).At the same time,the protein expression of survivin in the mulberry leaf leachate group decreased with the increasing concentration of the mulberry leaf leachate as compared with that of the control group.The results showed that the mulberry leaf leachate may inhibit proliferation of MDA-MB-231 cells,promote apoptosis,lead to G0-G1 arrest,and downregulate the survivin gene.
