Analysis of CARLo-5 and miR-219a expression levels in esophageal carcinoma tissues and their relationship with clinical characteristics and prognosis

Abstract:Objective To investigate the expression levels of cancer-associated region long non-coding RNA(CARLo-5)and mi-croribonucleic acid(miR)-219a in esophageal carcinoma(EC)tissues and their relationship with clinical characteristics and prognosis.Methods A total of 108 EC patients undergoing radical EC resection at the 943 th Hospital of Joint Logistics Sup-port Force of PLA from October 2020 to October 2021 were enrolled.Carcinoma tissues and adjacent normal tissues(>5 cm from the tumor margin)were collected using forceps after surgery.Differences in CARLo-5 and miR-219a expression between EC tissues and adjacent normal tissues were compared.Expression levels in EC tissues across different clinical characteristics were compared,and differences among patients with different prognoses at 3 years postoperatively were examined.Receiver operating char-acteristic(ROC)curves and area under the curve(AUC)were used to evaluate the predictive value of CARLo-5 and miR-219a for prognosis.Results Relative expression of CARLo-5 was significantly higher in EC tissues than in paracancerous normal tissues,while miR-219a expression was significantly lower(P<0.05).Significant differences in CARLo-5 and miR-219a expression were observed across pathological stages,invasion depths,and lymph node metastasis status(P<0.05).All 108 patients completed a 3-year follow-up:73 survived,35 died,with a cumulative survival rate of 67.59%(73/108).Patients were allocated to favorable prognosis(n=73)and poor prognosis(n=35)groups.The poor prognosis group showed higher CARLo-5 expression and lower miR-219a ex-pression than the favorable prognosis group,with statistical significance(P<0.05).ROC analysis revealed AUC values of 0.777(CARLo-5),0.746(miR-219a),and 0.832(CARLo-5 combined with miR-219a),indicating certain value in predicting the prognosis of EC patients.Using cutoff values from AUC analysis,high expression was defined as CARLo-5≥3.605 and miR-219a≥0.355.Kaplan-Meier curves showed the 3-year cumulative survival rate was 48.08%(25/52)in EC tissues with high CARLo-5 expression versus 85.71%(48/56)with low CARLo-5 expression(P<0.05),and 50.94%(27/53)in EC tissues with low miR-219a expression versus 83.64%(46/55)with high miR-219a expression(P<0.05).Conclusion CARLo-5 is upregulat-ed and miR-219a downregulated in cancerous tissues of patients with EC,correlating with pathological stage,invasion depth,and lymph node metastasis.Both biomarkers show value for predicting EC prognosis.
