The causal relationship between grain intake and insomnia:a Mendelian randomization study

Abstract:Objective To investigate the causal relationship between grain intake and insomnia using Mendelian randomization(MR).Methods Single nucleotide polymorphisms(SNPs)associated with grain intake were obtained from published genome-wide association studies(GWAS)and used as instrumental variables.The inverse-variance weighted(IVW)method was ap-plied to assess causality.Genetic heterogeneity was evaluated using Cochran's Q test,horizontal pleiotropy was tested via MR-PRESSO and MR-Egger regression,and sensitivity analysis was conducted using leave-one-out analysis.Results IVW analysis revealed that higher grain intake reduces the risk of insomnia(OR=0.91,95%CI:0.86-0.97,P=0.004),indicating a pro-tective effect.Each additional one-bowl weekly increase in grain intake was associated with a 9%reduction in insomnia risk.Conclusion A negative causal relationship exists between grain intake and insomnia.Increased grain consumption may modu-late sleep regulation and reduce insomnia risk.
