Research Progress on the Isolation Methods of Extracellular Vesicles

Abstract:Extracellular vesicles (EVs) are small vesicles released from various cells,which play an important role in cell to cell communication via transmitting proteins and genetic materials including DNA,mRNA and microRNA (miRNA) from original cell to target cell.EVs participate in many physiological and pathophysiological processes such as tissue regeneration,suggesting that EVs may be used as a biological marker for disease diagnosis.Currently,the frequently-used methods of EVs isolation were differential centrifugation,ultrafiltration and immunoaffinity.Several methods are always combined to increase the purity of isolated EVs,to stabilize their activities,and to reduce isolation time.Recent research reported that EVs were also isolated from semen and follicular fluid.It is necessay to explore the biological and clinical significance of EVs in semen and follicular fluid.The research progress on the isolation methods of EVs,and the advantage and disadvantage of each method were summarized in the present review as a reference for our EVs study.
