Abstract:Objective: To investigate the effect and mechanism of quercetin on human cervical cancer cell line SiHa. Methods:MTT assay was used to detect the effect of quercetin on proliferation of cervical cancer cell SiHa at different times (24 h, 48 h and 72 h) and different concentrations (0, 20, 40 and 80μmol/L);flow cytometry was used to detect the cell cycle in quercetin treated SiHa cells;RT-PCR and Western blotting was used to detect the expression of Cyclin D1 in quercetin treated SiHa cells. Results:The cell viability of SiHa cells by treating with quercetin for 24 h and 48 h in 40 μmol/L and 80μmol/L group were significantly lower than control group; The cell viability of SiHa cells by treating with quercetin for 72 h in 20μmol/L, 40μmol/L和80μmol/L group were significantly lower than control group;the number of G0/G1 cells in 20μmol/L, 40μmol/L and 80μmol/L group was significantly more than control group by treating with quercetin for 48 h;the protein level of Cyclin D1 was down-regulated in quercetin treated group. Conclusions: Quercetin could inhibit cell vitality and cell proliferation by down-regulating the expression of Cyclin D1 in cervical cancer cell SiHa.
