Effect of transformed recombinant ALA synthase gene (YHem1) on leaf photosynthetic energy conversion in strawberry
Abstract:In order to breed new varieties with higher photosynthetic capacity under stress conditions, the experiments were carried out to establish regeneration system from leaf explants of Benihonppe strawberry (Fragaria ananassa Duch.) and then a recombinant gene YHeml, yeast ALA synthase gene controlled by Arabidopsis thaliana HemAl gene promoter, was transformed into the detached leaves mediated with A- grobacterium tumefaciens. After kanamycin selection, PCR and RT-PCR detection, three plants were i- dentified to be the transgenic plants. The chlorophyll fast fluorescence showed that the transgenic straw- berry remained higher leaf maximal photochemical efficiency (F、/Fm), photosynthetic performance index (PI_ABS) and probability of a trapped exciton moving an electron into the electron transport chain beyond QA- (ψ.), while the approximate initial slope of the fluorescence transient (Mo) was lower in transgenie plants than in the wild type. This suggested that Yheml transformation might increase the capacity of photosyn- thetic energy conversion in strawberry leaves.
