Isolation and expression pattern of ACS and ACO genes in longan fruit

Abstract:In the present study, ACS and ACO genes from longan termed DlACS1 and DlACO1, were isolated using the combination of RT-PCR and RACE. DlACS1 protein was a polypeptide of 486 amino acids,including 7 conserved regions and 11 conserved amino residues,and DlACO1 protein consisted of 315 amino acids,covering 7 conserved regions and 9 conserved amino residues. Ethylene production exhibited a small ethylene peak at the early stage of fruit development,but decreased slowly thereafter and maintained a very low level at ripening stage. Northern blotting analysis showed that both DlACS1 and DlACO1 genes expressions in the pericarp were strongest at the rapid fruit growth stage,while in the aril, DlACS1 gene expression could be detected only at the early stage of aril development ,and DlACO1 gene expression declined gradually with aril growth,but slightly increased at ripening stage. In addition,the expressions of DlACS1 and DlACO1 did not exhibit tissue-specificity, but showing obvious difference in different tissues. These results provide basis for further study-ing the relationship between ethylene and the ripening of non-climacteric fruit.
