Study on the establishment of organogenesis regeneration system in vitro of Baihe-35-1, a line of Vitis pseudoreticulata

Abstract：Experiment was carried out with the leaves and stems of Baihe-35-1 grape cultivar for studying the effects of dif-ferent hormones on adventitious buds regeneration system so as to establish an adventitious shoot regeneration system in vitro. The main results were as follows: Different combination with hormones and proper explants can induce adventitious buds. A lower level in concentration of hormones was better for adventitious buds. Regeneration from stem was easier than from leaf. The combination of 1 mg·L~(-1) TDZ with 0.5 mg·L~(-1) NAA resulted in the highest frequency of adventitious bud production from leaf and stem with the rates of 3.3% and 12.8% ,respectively. The tissue culture plantlets were obtained from adventitious buds after being transferred onto the medium of MS +0.2 mg·L~(-1) IBA.
