A Study on Culture Conditions of Immature Embryo in Luotiantianshi Persimmon Variety

Abstract：The factors influencing immature embryo culture in Luotiantianshi persimmon variety (Diospyros kaki f.) were studied by Orthogonal design in 5 levels and 4 planes. The results showed that: (1) the optimum medium was MS(1/2N); (2) the optimum sucrose concentration was 7%; (3) activated carbon was the best for browning control; (4) the optimum IBA and Zeatin concentration was 0.3 mg/L and 0.44 mg/L，respectively; (5) the optimum kind of auxin and cytokinin was IAA and Zeatin; (6) a higher frequency of rooting occurred on half-strengthen MS(1/2N) medium supplemented with 1 mg/L IAA and dark treatment for 4 days.
