Impact and clinical implications of heart failure-induced myeloid-derived suppressor cell expansion on breast cancer progression

Abstract:Objective To analyze whether heart failure(HF)leads to an increase in myeloid-derived suppressor cells(MDSCs)in mice,and whether these expanded MDSCs are recruited into breast cancer tissues,thereby promoting tumor proliferation.Methods A mouse model of HF was established by isoproterenol(ISO)induction in mice bearing breast cancer.Cardiac function was assessed using M-mode echocardiography.Myocardial cell area was evaluated by hematoxylin-eosin(H&E)staining and wheat germ agglutinin(WGA)staining.The impact of HF on breast cancer development was assessed by comparing mammary tumor weight and volume,MDSC abundance,and their suppressive effect on T lymphocyte proliferation between the two groups.Results The tumor volume and weight in the ISO group were significantly greater than those in the control group(t=4.378 and 4.177,P<0.05).The content of MDSCs in the blood and tumor tissues of the ISO group was significantly higher than that in the control group,with a more pronounced increase in monocyte-type MDSCs within the tumor tissue(t=3.447,P<0.05).After co-culturing MDSCs recruited from the blood of HF mice with CD8+T lymphocytes,the proportion of proliferating CD8+T lymphocytes was 38.4%,indicating that MDSCs from HF mice significantly inhibited the T lymphocytes proliferation.Conclusions HF-induced expansion of MDSCs promotes breast cancer proliferation in mice,and HF-mediated alterations in immune function play a pivotal role in suppressing breast cancer progression.
