Association between knee osteoarthritis severity and tissue inhibitor of metalloproteinase-1,interleukin-1 and interleukin-6

Abstract:Objective:To analyze the the expression levels of tissue inhibitor of metalloproteinase-1(TIMP-1),interleukin-1(IL-1),and interleukin-6(IL-6)in patients with knee osteoarthritis(KOA)and their correlation with disease severity.Methods:A retrospective analysis was conducted on 97 KOA patients treated at Zhengzhou Orthopedics Hospital from January 2023 to March 2024(the KOA group).Another 68 people who underwent physical examination in the same hospital during the same period were selected as the healthy group.Serum levels of TIMP-1,IL-1 and IL-6 were measured,and their association with KOA severity were evaluated.Results:The KOA group showed significantly higher levels of TIMP-1(321.30±32.06 ng/mL vs.263.93±25.75 ng/mL),IL-1(226.50±35.06 pg/mL vs.47.01±6.25 pg/mL),and IL-6(29.89±4.86 pg/mL vs.4.19±0.42 pg/mL)than the healthy group(t=12.242,41.718,43.453;P<0.001).Among 97 KOA patients,levels progressively increased with disease severity:Mild group(n=32):TIMP-1=274.96±28.47 ng/mL,IL-1=160.45±26.06 pg/mL,IL-6=18.19±3.52 pg/mL;Moderate group(n=41):328.98±30.69 ng/mL,236.95±29.67 pg/mL,32.04±5.65 pg/mL;Severe group(n=24):369.96±52.36 ng/mL,296.74±31.09 pg/mL,41.85±6.97 pg/mL.(F=47.79,157.17,135.38;P<0.001).Positive correlations were observed between biomarker levels and KOA severity(r=0.607 for TIMP-1,r=0.663 for IL-1,r=0.715 for IL-6;P<0.001).Conclusions:Elevated serum TIMP-1,IL-1,and IL-6 levels are closely associated with KOA progression,suggesting their potential as biomarkers for disease monitoring.
