Expression and significance of midkine protein,proline-rich protein 11 and carbamyl phosphate synthase in patients with intrahepatic cholangiocarcinoma

Abstract:Objective:To analyze the expression and significance of midkine(MK)protein,proline-rich protein 11(PRR11)and carbamyl phosphate synthase(CPS1)in patients with intrahepatic cholangiocarcinoma(ICC).Methods:A total of 132 patients with ICC admitted to Zhengzhou Central Hospital from January 2020 to January 2022 were selected.The relationship between the expression of MK,PRR11 and CPS1 in cholangiocarcinoma tissues and pathological features was analyzed.Kaplan-Meier survival curve was used to analyze the relationship between MK protein,PRR11 and CPS 1 expression and the survival time of patients with cholangiocarcinoma.The single factors affecting the prognosis and death of ICC patients were analyzed,and the risk factors of ICC death were analyzed by COX risk regression.Results:The positive expression rates of MK,PRR11 and CPS1 in cholangiocarcinoma tissues were higher than those in tumor-adjacent tissues(P<0.05).The positive expression of MK,PRR11 and CPS1 in ICC patients with TNM stage Ⅲ to Ⅳ,moderately and poorly differentiated degree,lymph node metastasis,tumor length ≥ 3 cm and infiltration depth invading the outer membrane was higher than that in ICC patients with TNM stage Ⅰ to Ⅱ,well differentiated degree,no lymph node metastasis,tumor length<3 cm and no infiltration depth invading the outer membrane(P<0.05).The proportion of patients with TNM stage Ⅲ to Ⅳ,moderately or poorly differentiated,lymph node metastasis,positive expressions of MK,PRR11 and CPS1,tumor length ≥ 3 cm and invasion of the outer membrane in the event group(recurrence,metastasis or death of the disease during the follow-up)was significantly higher than that in the event-free survival group(P<0.05).Logistic analysis showed that TNM stage Ⅲ to Ⅳ,middle and low differentiation,lymph node metastasis,MK,PRR11,CPS1 positive,tumor length ≥ 3 cm and invasion depth into the outer membrane were independent risk factors for the death group of ICC patients(P<0.05).Conclusions:The proteins PRR11,CPS1,and MK are highly expressed in ICC tissues,and their expression levels are closely related to the clinical stage,pathological grade,and lymph node metastasis of ICC.The positive expression of these proteins is an independent risk factor for prognosis and mortality in ICC patients,providing certain reference information for predicting patient prognosis.
