Meta-analysis of rituximab in treatment of rheumatoid arthritis

Abstract:Objective:To systematically assess the clinical efficacy and safety of rituximab in treatment of rheumatoid arthritis.Methods:The relevant literatures of randomized controlled clinical study of rituximab (RTX) in treating rheumatoid arthritis were collected from the database of Pubmed,China national knowledge internet (CNKI),Wanfang Data and CQVIP database according to the key words such as ‘rituximab'and/or rheumatoid arthritis,the retrieval time was from building of the database to April 2016.The quality of the studies was evaluated,and Meta-analysis was performed with the software of RevMan 5.3.Results:A total of 5 studies (4 clinical trials were followed up at the 24th week and a clinical trial was followed up at the 52nd week) including 2 727 patients (1 240 cases treated with 1 000 mg rituximab,548 cases treated with 500 mg rituximab,and 939 cases served as placebo control) were enrolled.The Meta-analysis indicated that the efficacy of rituximab group was significantly superior to that of control group after a period of 24 weeks treatment [OR=2.78,95% CI (2.26,3.34),P<0.000 01].The incidence of side-effect had no statistical significance between the two groups [500 mg rituximab group:OR=0.91,95% CI (0.67,1.25),P=0.56;1 000 mg rituximab group:OR=1.00,95% CI (0.80,1.26),P=0.97].Conclusions:Rituximab is an effective drug in treatment of rheumatoid arthritis with no obvious difference in side-effect compared to that of placebo group.
