Research of the mechanism of epithelial mesenchymal transition during skin wound healing
Abstract:Objective:To explore the mechanism and significance of β2-adrenergic receptor (β2-AR) in the procedure of epithelial mesenchymal transition (EMT) during wound healing and tissue regeneration.Methods:The influence of β2-AR agonist (ISO) and inhibitor (ICI) on the expression of markers of mesenchymal transition (α-smooth muscle actin,vimentin,snail) or marker of epithelization (E-cadherin) in epidermis cells during wound healing and tissue regeneration was observed using western blot,reverse transcription polymerase chain reaction (RT-PCR) and immunofluorescence.Results:The epithelial cells gained mesenchymal phenotypic feature,and got stronger ability in movement and migration.β2-AR was a critical molecule which mediated EMT process.Conclusions:β2-AR is a key molecule in the procedure of EMT,and β2-AR mediated EMT is a biological basis of skin wound healing and matrix remodeling.
