The effects of ulinastatin on vasopermeability and water content of lung tissue in rats with scald injury

Abstract:Objective:To investigate the effects of ulinastatin on inflammatory mediators,vasopermeability and tissue water content of lung tissue after lethal scald inj ury in rats.Methods:Forty male Sprague-Dawley rats were randomly divided into scald group and ulinastatin group. A 50% total body surface area scald model was replicated in both groups by immersing the back,two limbs and abdomen of rats into 80°C water for 15,15,8 seconds re-spectively. Immediately after scald,rats were intravenously inj ected 1 ml of saline (scald group)or 1 ml of saline containing ulinastatin (40 000 U/kg,ulinastatin group ). Six hours and 1 day after scald,the plasma levels of tumor necrosis factor-α(TNF-α),interleukin (IL)-6 and IL-8 were measured by ELISA,the vasopermeability were detected by the method of Evans blue and the tissue water contents in lung and intestine were determined by the method of dry/wet weight.Results:Compared with scald group,the inflammatory mediators such as TNF-α, IL-6 and IL-8 in ulinastatin group were significantly lower than those in scald group 6 hours after inj ury,and the decrease was more significant 24 after scald (P<0.01). Compared with scald group,the visceral content of Evans blue and the tissue water content in lung were obviously lower in ulinastatin group than those in scald group,the difference was statistically significant at both time points(P<0.05 or P<0.01).Conclusions:The results indicate that ulinastatin can significantly alleviate systemic inflammation,inhibit visceral vasopermeability and tissue ede-ma,reduce blood loss from vessels,reduce the amount of fluid. Ulinastatin possesses potential value in early treatment for burn shock,and it improves the survival rate.
