Interleukin-13 in asthma and chronic obstructive pulmonary disease:from pathogenesis to targeted therapy

Abstract:Chronic respiratory diseases(CRDs),primarily bronchial asthma and chronic obstructive pulmonary disease(COPD),are characterized by chronic airway inflammation and remodeling.Their high morbidity and mortality rates present major challenges to global health.Interleukin-13(IL-13),a central mediator of the type 2 immunity,activate the JAK-STAT6 signaling pathway via the IL-4Rα/IL-13Rα1 receptor complex,or regulate in tumors and fibrotic diseases through IL-13Rα2.In the pathogenesis of asthma and COPD,IL-13 promotes pathological changes-including mucus hypersecretion,goblet cell hyperplasia,airway smooth muscle abnormalities,and epithelial barrier disruption via the JAK-STAT6 pathway.This review examines current states and future prospects of therapeutic strategies targeting IL-13,such as monoclonal antibodies and cell therapies,aiming to provide knowledge for targeted therapy.
