Research progress on antitubercular targets of marine natural products

Abstract:Tuberculosis(TB),an infectious disease caused by Mycobacterium tuberculosis(Mtb),presents challenges in clinical practice due to the emergence of severe drug resistance to first-line anti-TB drugs.Thus,it is an urgency for the development of novel anti-TB agents with new structures,targets,and mechanisms of action.Marine natural products serve as important resources for the discovery of anti-TB drugs.This review provides an overview of the current landscape of clinical anti-tuberculosis drugs under development and summarizes the progress in the research of anti-tuberculosis targets derived from marine natural products.For instance,the inhibition of shikimic acid kinase and mycobacterium protein kinase G disrupts the integrity of the cell wall of Mtb,and methionine aminopeptidase and protein tyrosine phosphatase A are involved in protein modification and degradation.In addition,protein tyrosine phosphatase B interferes with the signaling pathways by phosphorylating key signaling molecules of the host cell,thereby inhibiting Mycobacterium tuberculosis.Meanwhile,the novel anti-tuberculosis target acetohydroxyacid synthase(AHAS)catalytic subunit IlvB1 is specially emphasized,which holds significant promise for overcoming the current resistance bottleneck.With the continuous innovation in marine drug research and development technologies and the deepening of multidisciplinary collaborative research,marine natural products are exhibiting increasingly broad prospects in the field of anti-tuberculosis drug discovery.
