Effect of liposome-mediated transfection of protein kinase C-αantisense oligonucleotides on growth of CNE-2Z cells
Abstract:Objective:To observe the effect of protein kinase C-α(PKCα)antisense oligonucleotides transfected by cationic liposome(LP)on growth of CNE-2Z cells.Methods:PKCα antisense oligonucleotides were transfected by LP,and the cell growth was detected by MTT colorimetric assay.Results:(1)Cell growth index of CNE-2Z cells decreased as the strength of LP increased in the range of 2.000~8.000 mg*L-1(P<0.05),but there seemed to be no effect on cell growth with 1.000 mg*L-1 of LP.(2)Cell growth index reduced with a concentration increase ranging from 4.000 mg*L-1 to 16.000 mg*L-1 in antisense PKCα transfected by LP (P<0.001).Conclusion:It is indicated that antisense PKCα transfected with lower dose of LP may suppress growth of CNE-2Z cells,and this inhibitory effect shows a dose-effect relationship.
