Effects of quercetin on cell cycle and RNA polymerase activity in Hela cells

Abstract:Objective:To study the effects of quercetin on cell cycle and RNA polymerase in Hela cells.Methods:Hela cells were incubated with 4、20 and 100 μ mol/L quercetin for 3 days,respectively.Cell cycle was analyzed with flow cytometry,and the activities of total RNA polymerase and RNA polymerase Ⅱ were measured in isolated Hela cell nuclei by nuclear transcription in vitro.Results:After incubation with quercetin,the percentage of Hela cells at G1 phase increased in a dose-dependent manner,and the activities of total RNA polymerase and RNA polymerase Ⅱ were inhibited in the same way.Conclusion:Quercetin is able to arrest cell cycle progression of Hela cells at G1 phase,which may be related to the inhibition of the activities of total RNA polymerase and RNA polymerase Ⅱ.
