Effects of cryoprotectants on viability of cryopreservative corneal endothelial cells

Abstract:Objective:To investigate effects of osmotic stress of cryoprotectants on viability of cryopreservative corneal endothelial cells.Methods:New-born bovine corneas were divided into two groups randomly,and then cryopreserved with the simplified “two-step” method.Human serum albumin(HSA) and corneal-potassium-TES(CPTES) were chosen as cryoprotectant vehicles in group Ⅰ and group Ⅱ,respectively.The indexes,including cellular density,survival rate,polymegathism and pleomorphism,were compared respectively after thawing between two groups.Results:There were no significant differences on cellular density and survival rate between two groups,while group Ⅱ was superior to group Ⅰ on both polymegathism and pleomorphism.Conclusion:CPTES solution,an important ingredient of cryoprotectant,is helpful in improving the viability of cryopreservative corneal endothelial cells.
