The role of proteinase in the invasion process of human lung cancer cells

Abstract:To investigate the role of type Ⅳ collogenase,urokinase plasminogen activator(u-PA) and cathepsin B(CB) in the invasion process of high metastatic human lung cancer cell subline(PGCL3).Method:Three proteinases inhibitors,EDTA for type Ⅳ collagenase,trans-4-(aminomethy1)-cyclohexanecarboxylic acid(TA) for u-PA and N-ethylmaleimide(NEM) for CB,were chosen to observe their effects on the abilities of degrading and crossing the reconstructive matrigel of PGCL3 cells.The inhibitors were:inhibitor of cathepsin B).Results:Both TA and NEM could significantly inhibit the ability of invasion of PGCL3 cells,whereas EDTA appeared to have no inhibition. Conclusion:Both u-PA and CB are important enzymes in PGCL3 cell invasion,and the inhibition of u-PA and CB may be a prospective therapy of carcinoma.
