Therapeutic effect of Osthole on postoperative chronic pain in skin/muscle incision and retraction model rats based on gut microbiota

Abstract:Objective To investigate the therapeutic effect of intraperitoneally administered Osthole on postopera-tive chronic pain in a rat model of skin/muscle incision and retraction(SMIR),and to examine associated changes in gut microbiota.Methods Twenty-one SPF-grade male Sprague-Dawley rats were randomly divided into a blank group(Blank),a sham surgery group(Sham),and a model group(SMIR).Pain behavioral tests were conducted preoperatively and on days 1,3,7,and 14 postoperatively.On day 14 after surgery,fecal samples were collected for 16S rDNA sequen-cing to analyze gut microbiota composition.In a second set of experiments,21 SMIR model rats were equally randomized into three groups:a normal saline group(NS),a dimethyl sulfoxide group(DMSO),and an Osthole-treated group(OST).Pain behavioral assessments were performed at multiple time points before and after drug administration.On post-operative day 14,fecal samples were again subjected to 16S rDNA sequencing.Results Pain behavior scores were signif-icantly lower in the SMIR group compared to the Blank and Sham groups at all time points,indicating successful induction of chronic pain.Gut microbiota diversity was also reduced in the SMIR group.In contrast,the OST group showed allevia-ted pain behaviors and an increase in gut microbiota diversity compared to the NS and DMSO groups.Conclusion Intrap-eritoneal injection of Osthole can effectively reduce postoperative chronic pain in SMIR model rats.This analgesic effect may be associated with alterations in gut microbiota composition and diversity.
