Circulating MicroRNA in neoadjuvant chemotherapy for women with breast cancer

Abstract:Objective To investigate the correlation between circulating microRNAs(miRNAs)and the response to neoadjuvant chemotherapy(NAC)in women with breast cancer(BC).Methods A total of 263 BC patients undergo-ing NAC were enrolled.Peripheral blood miRNA levels were assessed at two time points:before initiating NAC(T1)and after the second chemotherapy cycle(T2).Changes in miRNA levels were analyzed for their correlation with chemothera-py response.Results Levels of four miRNAs(miR-145,miR-155,miR-195,and miR-21)showed significant decreases at T2 compared to T1(P<0.05).A reduction in miR-145 expression was negatively correlated with the effi-cacy of NAC in Luminal B-type BC patients(P<0.05).Furthermore,reduced miR-145 expression was associated with a lack of pathologic complete response(pCR)in ERBB2-positive BC patients following NAC(P<0.05).Conclu-sion Peripheral blood miR-145 can serve as a predictive biomarker for NAC response in BC patients.Clinicians may u-tilize this marker for early intervention or tailored therapeutic strategies.
