Specificity of gut microbiota distribution in adolescent MAFLD rats with different glucose and lipid levels

Abstract:Objective To investigate the distribution differences of gut microbiota in adolescent rats with metabol-ic associated fatty liver disease(MAFLD)under varying glucose and lipid levels.Methods Thirty-two 6-week-old specific pathogen-free(SPF)male SD rats were randomly assigned to four groups:control(C)group,model(S1)group,high sugar high fat(M)group,and exercise intervention(S5)group,with 8 rats per group.The control group was fed standard chow,while the other three groups were given a high-fat diet.After 8 weeks on a high-fat diet,the High Sugar High Fat group received a single intraperitoneal injection of streptozotocin(35 mg/kg×body weight),while the Exercise Intervention group started a nightly swimming regimen at 18:00.At the end of the 14th week,rats were euth-anized,and their gut microbiota distribution,body weight,blood glucose,transaminase,and lipid levels were measured.Results There were statistically significant differences between the normal control group and the high glucose and high lipid group and the model group(P<0.01),and there was a statistically significant difference between the exercise inter-vention group and the high glucose and high lipid group and the model group(P<0.01),while there was no difference between the normal control group and the exercise intervention group(P>0.05).Cholesterol and triglycerides in the nor-mal control group showed significant differences compared to the other three groups(P<0.05).Alpha diversity analysis of the four groups showed that the species abundance and diversity of the normal control group were higher than those of the other three groups.The relative abundance of Staphylococcus and Corynebacteria in the exercise intervention group was significantly increased.Conclusion Exercise intervention promoted advantageous bacterial species,potentially improving glucose and lipid levels in adolescent SD rats on a high-fat diet.
