Circulating inflammatory markers and prostate cancer risk:a Mendelian randomization study

Abstract:Objective To investigate the genetic causal relationship between circulating inflammatory markers and the risk of prostate cancer using a two-sample Mendelian randomization(MR)approach.Methods Data on circu-lating inflammatory markers and prostate cancer were obtained from the largest available genome-wide association study(GWAS)in 2021.Significant single nucleotide polymorphisms(SNPs)were selected as instrumental variables.Five MR analysis methods,including inverse-variance weighting(IVW)as the primary method,were applied to assess the causal relationship between circulating inflammatory markers and prostate cancer risk.SNP horizontal pleiotropy and heterogeneity were explored using the MR-Egger intercept test,MR-multivariate residual and outlier method,and Cochran's Q test.Leave-one-out analysis was performed to investigate the influence of each SNP on MR results.Results IVW analysis indicated that CTACK(odds ratio[OR]:1.15,95%confidence interval[CI]:1.03-1.27,P=0.012)is a risk factor for prostate cancer,while IL-2ra(OR:0.90,95%CI:0.81-1.00,P=0.045)and IL-12p70(OR:0.87,95%CI:0.76-0.99,P=0.038)are protective factors.Conclusion This study provides new evidence for the causal relation-ship between inflammatory markers and prostate cancer.It lays the foundation for further research into the mechanisms of prostate cancer development and progression,and may help in developing more effective therapeutic strategies,improving clinical outcomes.
