Effect of Ruscogenin regulation on DJ-1 expression on apoptosis in endometrial cancer Ishikawa cells

Abstract:Objective To investigate the effect of the active component of traditional Chinese medicine,ruscoge-nin,on the expression of DJ-1 and apoptosis in endometrial cancer Ishikawa cells.Methods Endometrial cancer Ish-ikawa cells were used as the research objects.The CCK-8 method was used to analyze the effect of ruscogenin on the pro-liferation of Ishikawa cells.qRT-PCR was used to detect the expression of DJ-1 mRNA in Ishikawa cells treated with ruscogenin,and flow cytometry was used to detect the effect of ruscogenin on apoptosis in Ishikawa cells.Results After treatment with different concentrations of ruscogenin,the inhibition rate of cell proliferation in Ishikawa cells was signifi-cantly increased compared to the control group,showing a time-concentration-dependent manner.After treatment with ruscogenin for 48 hours,the IC50 value of the inhibition rate was 78μmol/L.Compared with the control group,the expres-sion level of DJ-1 mRNA in Ishikawa cells treated with 78μmol/L ruscogenin for 48 hours was significantly decreased(P<0.05),while the early apoptosis rate(P<0.01)and late apoptosis rate(P<0.05)were significantly increased.Conclusion Ruscogenin can promote apoptosis in endometrial cancer Ishikawa cells by downregulating the expression of DJ-1,and this effect is dose-dependent.
