Effects of Realgar on proliferation,apoptosis and HPVE6/IL-6/STAT3 pathway in cervical cancer cells

Abstract:Objective To investigate the impact of Realgar on proliferation and apoptosis of cervical cancer cells and investigate its relationship with the E6 and IL-6/STAT3 pathways.Methods The CCK-8 assay was used to as-sess the effects of different concentrations(0,25 μg/mL,50 μg/mL,75 μg/mL,100 μg/mL)of Realgar on the prolif-eration of cervical cancer HeLa and Caski cells.Flow cytometry was employed to detect apoptosis in HeLa and Caski cells.Western blot examined the influence of various concentrations of Realgar on the expressions of STAT3 and p-STAT3 pro-teins in both cell lines.qPCR was used to measure the gene expression levels of E6,IL-6,PARP,and Bcl-xL.Results Realgar significantly inhibited the proliferation of both cervical cancer cell lines and promoted apoptosis,show-ing a certain concentration dependency.Western blot and qPCR results indicated downregulation(P<0.05)of STAT3,p-STAT3,E6,IL-6,PARP,and Bcl-xL expression,with a concentration-dependent trend.Conclusion Realgar can inhibit the proliferation and promote apoptosis of HeLa and Caski cells,potentially through the IL-6/STAT3 pathway mediated by E6.
