NLRP3 inflammasome regulates bacterial eradication during Pseudomonas aeruginosa infection

Abstract:Objective To investigate the role of NLRP3 inflammasome in the immune response of macrophages against Pseudomonas aeruginosa (PA) infection.Methods ELISA, Real-time PCR and Western blot assay were used to assess the expressions of IL-1β, caspase-1 and NLR (mainly NLRP3) during PA infection. Macrophages were transfected with siRNA to silence the components of NLRP3 inflammasome. ELISA assay was used to determine IL-1β and caspase-1 secretion. Immunofluorescence staining and confocal microscopy were applied for observation of NLRP3 inflammasome. Bacterial plate count assay was carried out to determine the PA clearance. Real-time PCR, Griess reaction, flow cytometry, Western blot and immunofluorescence experiments were used to assess the antimicrobial peptides, NO, ROS and autophagy. Rapamycin or siRNA were used to block or induce autophagy, respectively. Results NLRP3 inflammasome was activated after PA infection. NLRP3 inflammasome reduced bacterial clearance and autophagy during PA infection. Silencing NLRP3 inflammasome induced PA clearance and autophagy, while had no effects on the production of antimicrobial peptides, NO or ROS.Conclusion The activation of NLRP3 inflammasome in response to PA infection impairs bacterial eradication via autophagy.
