The safety analysis of PPP2R5C-siRNA

Abstract:Objective To analyze the safety of PPP2R5C small interference RNA. Methods Micromorphological changes between the cells transfected with PPP2R5C small interference RNA 72 h K562 cells and MOCK K562 cells by atomic force microscope(AFM). The expression of PPP2R5C gene in K562 cells, healthy individual bone marrow mononuclear cells(MNCs), MNC-siRNA and MOCK-siRNA were detected by real-time PCR. The effect of PPP2R5C-siRNA on the differentiation and proliferation ability of healthy individual BM-MNCs in vitro was analyzed by methylcellulose semi-solid culture. Results There was no significant difference in cell height and radius among the cells transfected with PPP2R5C small interference RNA. The expression of PPP2R5C in BM-MNCs of 3 healthy individuals was significantly lower than that of K562(P < 0.05). There was no significant change in PPP2R5C gene expression in BM-MNCs before and after transfection. There was no significant difference in the formation of BFU-E, CFU-GM and CFU-Meg in healthy human BM-MNCs after PPP2R5C-siRNA transfection and seed plate 14 d compared with MOCK control group(P = 0.723, 0.826, 0.448). Conclusion PPP2R5C-siRNA has no significant effect on the proliferation and differentiation of BM-MNCs in healthy individual in vitro, which preliminarily illustrates its safety.
