Effects of alpha-enolase on epithelial-mesenchymal transition of human MGC-803 gastric cancer cells

Abstract:Objective To investigate the effect of overexpression of α-enolase (ENO1) on epithelial -mesen-chymal transition (EMT) in gastric cancer cell line MGC -803.Methods The eukaryotic expression vector pcDNA3. 1/ENO1 was constructed and sequenced when the coding region of ENO 1 gene was augmented, and then transfected into MGC-803 cells as ENO1 group.At the same time, empty plasmid pcDNA3.1 was transfected as the control group .The expression of ENO1 mRNA and protein in each group was observed using Real -time PCR and Western Blot .Expression levels of the EMT specific marker genes , such as E -cadherin, Snail and Vimentin, were compared between the two groups after 24-hour transfection.The siRNA of ENO1 was synthesized and transfected to cancer cells by liposome .Real-time PCR was used to detecting the expression of EMT genes between the interference group and control group after 24 hours transfection.Results Real-time PCR results showed that the expression of ENO 1 mRNA in ENO1 group was sig-nificantly higher than control group ( P <0.05).Western Blot showed that the relative expression of ENO 1 protein in ENO1 group was also significantly higher than control group (P<0.05).The expression of E-cadherin, Snail or Vimen-tin in ENO1 group showed no significant difference from the control group (P>0.05).The expression of E -cadherin, Snail and Vimentin in interference group were not significantly differed from the control group at 24 hours ( P>0.05). Conclusion Overexpression and interference of ENO 1 in gastric cancer cells has no obvious effect on EMT.
