Establishment and evaluation of cholestasis hepatic fibrosis rhesus monkey model induced by complete bile duct obstruction

Abstract:Objective To establish the cholestasis hepatic fibrosis rhesus monkey model induced by bile duct obstruction. Methods A total of 6 healthy Chinese experimental rhesus monkey were selected. Complete bile duct ligation was performed on all 6 subjects. Continuously observation was carried out for 90 days after operation. The jaundice and liver fibrosis were observed. Liver fibrosis was assessed using ultrasound 1 month and 3 months after operation, as liver cirrhosis and liver function indexes were assessed before and 3 months after operation. Liver biopsy and abdominal examination were performed 4 months after operation. Results Three months after complete biliary obstruction, jaundice was severer as prolonging time, and significant biliary dilatation and uneven liver parenchyma changes were observed 1 and 3 months after operation. Compared with preoperative data, increased liver cirrhosis was observed 3 months after operation. Serum total bilirubin (TBIL) and alanine aminotransferase (ALT) levels were significantly increased 2, 4 and 6 weeks after operation (P < 0. 05). Four months after operation, the liver pathological examination results showed that there were a lot of fibrous bands in the liver with the main area in the portal area, and laparotomy results revealed liver swelling and uneven surface. Conclusion It is effective to establish a stable and reliable hepatic fibrosis animal model with rhesus monkey by the method of complete bile duct obstruction.
