Evaluating the effect of pioglitazone on inflammation and calcification in rabits using 18F-FDG-PET/CT

Abstract:Objective To evaluate effects of pioglitazone on calcification and inflammation in rats. Methods Twenty male New Zealand white rabbits were randomly divided into two groups, pioglitazon group (n = 10) and control group (n = 10). Atherosclerosis was induced in all rabbits. The rabbits in pioglitazone group received pioglitazone (10 mg·kg-1·d-1) daily. Mid-stage PET/CT scan and end-stage PET/CT were performed at the 8th and 18th week. The standardized uptake values (SUVs) were measured and documented. Finally, all rabbits were euthanized, and aortic histopathological assessment and correlation analyses were performed. Results At the end of the 8th week, no significant in SUV was observed between the 2 groups. At the end of the 18th week, the SUV was significantly reduced in pioglitazone group, with significant difference between the 2 group. Histopathological examination of aortas in the control and pioglitazone groups showed significantly lower macrophage density (12. 31 ± 6. 20% vs. 6. 27 ± 3. 37%, P < 0. 01) and tissue calcification area (6. 19 ± 3. 48% vs. 5. 04 ± 3. 20%, P < 0. 05) in pioglitazone group. At the end of the 18th week, macrophage density and tissue calcification area were positively correlated with the SUV. Conclusion Pioglitazone improves the early vascular microcalcification and inflammation.
