The effect of Jiawei guizhifulingwan on Wistar rats with gouty arthritis

Abstract:Objective To study the effect of Jiawei guizhifulingwan on Wistar rats with gouty arthritis, and to clarify the its optimal concentration and mechanism. Methods Thirty-six male Wistar rats were randomly divided into 6 groups, blank group, model group, colchicine group, and high-, medium-and low-dose groups. The degree of joint swelling, serum uric acid, pathological inflammatory state of the soft tissues, the immunohistochemical expression of myeloid differentiation factor 88 (MyD88) and interleukin-1m (IL-1β) were observed. Results The rats in the blank group had no swelling, normal serum uric acid level, no inflammatory cell infiltration in the soft tissues, and normal expression of IL-1ll infiltration in the soft ti. Compared with the blank group, higher serum uric acid, obvious joint swelling, obvious pathological gout, and significant higher expression of IL-1 higher rouption iobserved in model group. Compared with the model group, each index was lower in the 3 drug groups, though they were slightly higher than the blank group. The indexes in high-dose group were similar with colchicine group. Conclusion Jiawei guizhifulingwan has a certain therapeutic effect on Wistar rats, and it is most prominent in high dose group. It may be achieved by inhibiting the expression of IL-1β and MyD88 in the synovial tissue of the joint.
