Study on the induced apoptosis of cholangiocarcinoma QBC939 cells by Osthole

Abstract:Objective To study the effects of Osthole on cholangiocarcinoma cells and its mechanism. Methods Using cell culture, MTT assay, Giemsa staining, Hoechst33258 fluorescence staining, AO/EB staining, flow cytometry and Western blot methods, the proliferation and morphological changes of cholangiocarcinoma cells, apoptosis rate, expression change cycle and apoptosis related protein of cholangiocarcinoma QBC939 cells treated by osthole were investigated. Results The results showed that proliferation of human cholangiocarcinoma QBC939 cells was inhibited by osthole with dose-and time-dependent manners, with the IC50 values of 0. 169 63 mmol /L and 0. 100 19 mmol /L for 24 h and 48 h, respectively. The apoptosis rate reached 37. 15% after treatment with osthole for 48 h, and morphological changes in characteristics of cell apoptosis were detected. Meanwhile, the expression level of apoptosis-related proteins Fas and Caspase-3 were changed after exposed to osthole. Conclusion Osthole has a good anti-cancer effect, which may be via the exogenous apoptosis pathway of caspase.
