Abstract:Objective To investigate the feasibility of using carbon dots as a marker of sinapine thiocyanate, and to clarify its molecular mechanism. Methods During the cell culture experiment, the intervention of the carbon dots and carbon dots labeled sinapine thiocyanate were performed respectively. The imaging effect of the sinapine thiocyanate labeled with carbon dots was observed by confocal laser scanning technique. The cytotoxicity of carbon dots and labeled sinapine thiocyanate were examined by CCK-8 method. Results HaCaT cell line was cultured with confocal dishes. Using blank carbon dots as the control, the thiocyanate labeled with carbon dots was used as the experimental sample, and the green fluorescence was observed by confocal laser scanning microscope. With the increase of culture time, the fluorescence signal increased gradually. This indicated that the intake of mustard and thiocyanate was more and more important in HaCaT cells. Conclusion The carbon dots can be used as an effective marker for sinapine thiocyanate, which will help to reveal the molecular mechanism of sinapine thiocyanate.
