Protective effects of astragaloside in rats with sepsis

Abstract:Objective To investigate the protective effects of astragaloside in septic rats. Methods Fifty SD rats were randomly divided into Sham-operation group, cecal ligation and puncture-18h group ( CLP-18h) , cecal ligation and puncture-72h group ( CLP-72h) , astragaloside-18h group ( AST-18h) , astragaloside-72h group ( AST-72h) . Except for those in Sham group, the rats in the other groups received cecal ligation and puncture method to induce sepsis. The rats in CLP groups received intragastric administration of NS ( 10 mL/kg) after CLP. Astragaloside were intragastric administrated in the same time points. Alanine transaminase ( ALT) , aspertate aminotransferase ( AST) , total bilirubin( TBIL) , creatinine ( Cr) , urea, creatine kinase-MB ( CK-MB) , lactate dehydrogenase ( LDH) , and alpha-hydroxybutyric dehydrogenase ( α-HBD) were detected by biochemical analyzer. Blood gas analyzer was used to measure related indicators. The levels of complement 5a, mobility group box-1 protein B1 ( HMGB-1) , interleukin 6 ( IL-6) , interleukin 6 ( IL-10) , and myeloperoxidase ( MPO) were analyzed by enzyme-linked immune-absorbent assay. Hematoxylin-eosin ( HE) staining was used to observe the histopathological changes of heart, lung and intestine. The spleen index and thymus index were tested. Results The levels of Cr, CK-MB, LDH and α-HBD in serum were reduced after treatment with astragaloside. The levels of pHTC, iCa ++ and FHHB were changed after treatment with astragaloside. The levels of IL-6, HMGB-1 C5a and MPO in serum were reduced and the level of IL-10 was increased. On morphological aspects of the tissues, the injury of heart, lung and intestinal tissue were significantly improved. HMGB-1 was positively correlated with IL-6 and IL-10. MPO was positively correlated with IL-6, HMGB-1 and IL-10. Thymus index was negatively correlated with IL-6 and spleen index, the difference was statistically significant ( P<0. 05) . Conclusion Astragaloside can inhibit the inflammatory infiltration; reduce inflammation on the heart, lung and intestinal tissue damage; thus protects rats from sepsis.
