The research of lanthanum chloride-induced reverse of cisplatin resistance in ovarian cancer cells

Abstract：Objective To investigate the effect of lanthanum chloride -induced reverse of cisplatin resistance in ovarian cancer cells , and to determine the target concentration of lanthanum chloride .Methods The COC1 and COC1/DDP cells were cultured with different concentrations of lanthanum chloride and cisplatin co -culture.Cell proliferation and inhibition rates were assessed by Trypan Blue Staining and MTT assays , respectively.Apoptotic morphological chan-ges were observed by transmission electron microscope ( TEM ) .Results Under different concentrations of lanthanum chloride and cisplatin co -culture , the cell proliferation was significantly inhibited and apoptosis was promoted in COC 1 and COC1/DDP cells by high concentration lanthanum chloride .Low concentration lanthanum chloride had no influence on COC1 or COC1/DDP cells, neither with cisplatin co-culture.Lanthanum chloride at 1.5 μmol/L alone had no effect on COC1 or COC1/DDP cells, but with cisplatin co -culture, proliferation inhibition and apoptosis promotion were ob-served in COC1 and COC1/DDP cells.Conclusion Lanthanum chloride at 1.5 μmol/L can reverse cisplatin resistance in ovarian cancer cells , while lanthanum chloride over 1.5 μmol/L inhibits the ovarian cancer cells with cisplatin resist-ance .
