The effects of simvastatin on serum lipid metabolism and the liver oxidative stress during early sepsis in rats

Abstract:Objective To study the effects of simvastatin on serum lipid metabolism and the liver oxidative stress in rats with lipopolysaccharide (LPS) -induced sepsis.Methods Fifty-six male Sprague -Dawley (SD) rats were randomly divided into three groups, control group (n=8), model group (n=24) and simvastatin group (n=24).The septic model was set up in rats receiving intra -peritoneal injection of LPS (10 mg/kg).The control group or model group were consecutively given with saline (1 mL/d) intra-gastrically for one week before intra -peritoneal injection of saline or LPS, respectively.Simvastatin group was given with simvastatin (20 mg? kg-1? d-1) intra-gastrically for one week. The levels of serum total cholesterol ( TC) ,, triglyceride ( TG) ,, low density lipoprotein ( LDL) , high-density lipopro-tein (HDL) and liver malondiadehyde (MDA), and the activity of liver superoxide dismutase (SOD) were assessed 6, 12 and 24 hours after modeled .Results Compared with control group , serum TC, TG, and LDL levels and liver MDA content were firstly increased and then decreased in model group , while the activity of liver SOD was significantly de-creased in model group (P<0.05).Compared with model group, the serum TG was significantly reduced at the 6th hour in simvastatin group (P<0.05), serum TC was significantly reduced at the 24th hour (P<0.05), liver MDA content was significantly reduced at the 6th and the 12th hour (P<0.05), and the activity of liver SOD showed no significant difference between the 2 groups.Conclusion Simvastatin reduces serum total cholesterol level , but it can reduce oxidative stress -induced liver injury during early sepsis in rats .
