A screening for lncRNAs related to RSV-induced asthma in dendritic cells

Abstract：Objective To screen the differentially expressed long non -coding RNAs ( lncRNAs ) in dendritic cells (DCs) in respiratory syncytial virus (RSV)-induced asthma, and to study the role of lncRNAs in RSV -induced asthma.Methods We set up three groups , HDC -BEAS +RSV group, HDC -RSV group and HDC -CONTROL group.Human bronchial epithelial cell BEAS -2Bs were infected with RSV and immature DCs were co -cultured by Tran-swell in HDC-BEAS+RSV group .Immature DCs were infected with RSV in HDC -RSV group .Immature DCs were in-fected with culture medium without RSV in HDC -CONTROL group.The differentially expressed genes and lncRNAs of DCs were subsequently screened by Affymetrix Human Transcriptome Array 2.0, and cluster analysis , GO analysis and Pathway analysis were conducted .Results There were 25 genes and 29 lncRNAs with up-or down-regulation in HDC-BEAS+RSV group comparing with controls , among which, only 5 genes and 2 lncRNAs were down-regulated.The up-regulated genes were enriched in pro -inflammatory immune reaction such as signal transduction , chemokine signaling and cytokine-cytokine receptor interaction .Conclusion We have screened out the differentially expressed lncRNAs in RSV -induced asthma .And it would contribute to the further study for the role of lncRNAs in RSV -induced asthma .
