Abstract:Objective To explore the effects of miR-135b on HOXA10 in endometrial cancer. Methods The expression of miR-135b and HOXA10 in human endometrial cancer tissues and their para-carcinoma tissues was assessed by real-time PCR.miR-135b mimics (miR-135b up-regulated expressions) and miR-135b inhibitors (miR-135b down-regulated expression) were transfected into human endometrial cancer RL-952 cells by liposome method. The transfection efficiency was detected by RT-PCR.Wound healing assay was performed to detect the cell migration ability.Cell proliferation and cell cycle were analyzed 48h after transfection by flow cytometry. The expression of HOXA10 mRNA and protein were assessed by RT-PCR and Western blot. Results The expression of miR-135b in human endometrial cancer tissues was significantly higher than their para-carcinoma tissues (P<0.05).Expression of HOXA10 in endometrial carcinoma tissues was significantly lower than their para-carcinoma tissues (P<0.05).Compared with NC group, the mRNA and the protein expression of HOXA10 was significantly down-regulated in miR-135b mimics group (P<0.05).However, in miR-135b inhibitors group, the mRNA and the protein expression of HOXA10 was significantly up-regulated (P<0.05).MiR-135b can promote the proliferation of endometrial cancer cells (P<0.05).Conclusion MiR-135b plays an important role in the occurrence and development of endometrial carcinoma by regulating HOXA10.
