The diagnostic application of plasma lncRNA-HIT in breast cancer

Abstract:Objective To measure the expression of lncRNA-HIT in plasma of breast cancer patients and dis cuss its potential value as a biomarker in diagnosis of breast cancer.Methods The plasma samples were collected in 124 cases of breast cancer,59 cases of benign breast tumor and 48 normal women.The expression of HIT was assessed by quantitative real-time reverse transcription polymerase chain reaction (real time RT-PCR).Simultaneously,the corre lation between the expression of HIT and the clinicopathological features of patients was analyzed.Receiver operating characteristic (ROC) curve was performed to evaluate the diagnostic values of plasma HIT in breast cancer patients from controls.Results The plasma expression level of HIT in breast cancer [0.12 (0.02,0.33)] was significantly lower than the control group [0.55 (0.20,1.59)] (U =1 020,P < 0.000 1).The plasma expression level of HIT in breast benign lesion group [0.10(0.02,0.19)] was significantly lower than the control group (U =308.5,P <0.000 1).The HIT expression in breast cancer cases was lower than that of benign lesion group (U =3 028,P =0.060 2).Low expression of HIT was significantly correlated with estrogen receptor (ER) (P =0.021) and lymphatic metastasis (P =0.033).The area under the ROC curve (AUC) of plasma HIT was 0.799,with sensitivity and specificity of 83.3% and 67.7%,respetively.Conclusion Low expression of HIT in breast cancer may be a potential diagnosis biomarker for breast cancer.
